- Course objectives
- The main goals of the subject are understanding the spectroscopic, chromatographic, electromigration, and physical methods of biochemical analysis.
- Learning outcomes
- At the end of this course, the students will be able to theoretically understand and practically apply the spectroscopic, chromatographic, electromigration, and physical methods of analysis in biochemistry. They will able to apply this knowledge in their scientific research, working on bachelor, master, and PhD theses and finally also during their following employment.
- Syllabus
- 1. Electronic spectra of molecules, transitions, ground and excited state, influence of the environment, UV-Vis spectroscopy, use to determine substances and study the structure of proteins.
2. Chiroptical methods and their principle. ORD and CD and their use to study protein conformation.
3. IR spectroscopy and its use to study protein structure, Raman scattering and its use to study protein structure.
4. Atomic absorption and atomic emission spectroscopy.
5. Luminescence methods. Quantum yield, influence of the environment, use to determine substances and study protein conformation, fluorescence quenching, energy transfer.
6. Microscopy. Methods of optical microscopy, electron microscopy, and scanning probe microscopy.
7. Immunochemical methods. Preparation and use of antibodies in bioanalytical chemistry, formats of immunochemical assays.
8. Biosensors. Biocatalytic and bioaffinity sensors, the most important types of transducers.
9. Chromatographic methods, principles (adsorption, ion-exchange), chromatofocusing.
10. Electromigration methods, principles, zone electrophoresis, IEF, isotachophoresis, blotting.
11. Mass spectrometry. - Teaching methods
- Lectures with a demonstration of the taught topics by means of demonstration videos and programs.
