Course objectives
The aim of this course is to give undergraduate and PhD students a basic orientation in principles of morphological, cytogenetic, and molecular methods currently used in zoological research. After finishing the course, students should gain theoretical knowledge and practical skills in the following topics: Analysis of phenotype (epigenetic traits, quantitative traits, geometric morphometrics); Cytogenetics (karyotype analysis, banding, FISH, "chromosome painting“); Protein and DNA electrophoresis; DNA analysis (DNA isolation, DNA-DNA hybridization, PCR, real-time PCR, fingerprinting, RAPD, AFLP, analysis of microsatellites, LINE, SINE, and SNP; sequencing, "next generation sequencing, RFLP, DGGE, TGEE, SSCP, cloning, microarrays); Methods of analysis (population-genetic data: analyses based on allele frequencies vs. "individual-based" models; phylogenetic data: distance methods, maximal parsimony, maximal likelihood, Bayesian analysis).

  • 1. Analysis of phenotype (signal phenotypes, epigenetic traits, quantitative traits, landmark analysis)
  • 2. Cytogenetics (karyotype analysis, banding, FISH, „ chromosome painting“) and elektroforetic methods (proteins, DNA)
  • 3. DNA aAnalysis I (DNA isolation, genetic markers - nuclear vs. cytoplasmic DNA, DNA-DNA hybridization, PCR, real-time PCR, "multi-locus" DNA markers: fingerprinting, RAPD, AFLP)
  • 4. DNA analysis II ("single-locus" DNA markery: mikrosatelity, LINE, SINE, SNP)
  • 5. DNA analysis III (sequencing methods, "next generation sequencing") 6. DNA analysis IV (SNP and their analysis: RFLP, DGGE, TGEE, SSCP, cloning, microarrays)
  • 7. Methods of analysis I (population-genetic data) - analyses based on allele frequencies vs. "individual-based" models
  • 8. Methods of analysis II (phylogenetic data) - distance methods, maximal parsimony, maximal likelihood, Bayesian methods
  • 9. Practical class I (chromosomal preparations, karyotype analysis)
  • 10. Practical class II (protein electrophoresis)
  • 11. Practical class III (PCR, real-time PCR)
  • 12. Practical class IV (analysis of DNA fragments after PCR, electrophoresis, fragment analysis, capillary SSCP)
