Course objectives
At the end of the course students should be able to:
understand molecular principles of origin and activity of reactive oxygen and nitrogen species;
explain their physiological role in animal organism;
review the role of free radicals in diseases;
categorize defence reactions against free radicals;
understand principles of methods evaluating oxidative damage.
Syllabus
  • Biology of oxygen (Oxygen and evolution of life on Earth; Oxygen cycle in nature; Photosynthesis; Respiratory chain). Chemistry of free radicals (Reactive oxygen species; Role of transition metal ions; Nitric oxide and derived molecules). Sources of free radicals (Endogenous physiological or pathological processes: Leak of electrons from metabolic pathways, Peroxisomes, Xanthine/xanthine oxidase, Metabolism of arachidonic acid, Autooxidation of catechols and flavonoids; Exogenous sources, Food constituents, Drugs, UV irradiation, Ionising irradiation, Polluted environment). Physiology of free radicals (Phagocytosis; NADPH oxidase of professional phagocytes; Microbicidal mechanisms based on oxygen; Chronic granulomatous disease (CGD); Gene regulation; Apoptosis; Oxidative burst in plants as defence mechanism against infection). Role of free radicals in diseases (Lipid peroxidation; Protein degradation; DNA damage; Regulation of oxidative stress). Defence against free radicals (Prevention of free radical generation; Compartmentalisation of free radicals; Free radical scavengers; Antioxidant defence and damage repair; Antioxidant pharmacological intervention). Pathology of free radicals (Oxidative stress and aging; Reproduction; Ischemia-reperfusion injury; Inflammation; Atherosclerosis; Brain dysfunction; Cancer; Myocarditis; Diabetes mellitus; Eye diseases; Adult respiratory distress syndrome (ARDS); Oxidative injury to lungs induced by xenobiotics; Radiation injury; Rheumatoid arthritis; Skin injury). Methods of oxidative damage evaluation (Detection of free radicals; Evaluation of antioxidant activity of biological samples; Determination of damage to biological macromolecules).
