Course objectives
At the end of this course students should be able to understand and to explain:
basic concepts in toxicology and chemical safety;
adverse local, acute and chronic systemic and late effects, dangerous physical-chemical properties and ecotoxicity of substances and preparations;
methods and guidelines of testing, assessment, classification and labelling of dangerous substances and preparations;
factors determining the adverse effects of substances on human health, interaction of substances with organism - exposure, absorption, distribution, biotransformation and elimination of xenobiotics, molecular, cellular and organ levels of interaction;
Czech and EU legislation on substances and preparations, public health and occupational safety and health;
occupational exposure limits, maximum permissible concentrations of substances in water, maximum residue levels in foods;
principles of the risk analysis and the protection of workers from the risk related to exposure to chemicals, emergency measures in cases of accidental chemical exposure;
dangerous properties of important inorganic and organic substances;
sources of information on dangerous properties of substances and preparations.
Syllabus
  • 1. Introduction, aims and programme of course.
  • Basic concepts - toxicology, chemical safety, harmful chemicals, xenobiotics, exposure, dose, effect, latent period, response, hazard, risk, classification, dangerous substances and preparations, danger symbols, risk and safety phrases.
  • Local effects of substances, testing and evaluation of acute irritation and corrosion, irritant and corrosive substances and preparations.
  • 2. Systemic effects, acute and chronic intoxications, testing of acute, subacute, subchronic and chronic toxicity of chemicals, LD50, LC50, NOAEL, LOAEL, evaluation of toxicity of substances, very toxic, toxic and harmful substances and preparations.
  • Sensitisation, allergy, testing and evaluation of sensitisation of chemicals, sensitising substances and preparations.
  • 3. Late effects of substances.
  • Mutagens, gene mutations, structural and numerical chromosome aberrations, genetic toxicology, bacterial reverse mutation tests, mammalian bone marrow chromosome aberration test, DNA damage and repair tests, epidemiological studies, evaluation of mutagenicity of chemicals, mutagenic substances and preparations.
  • 4. Carcinogenicity, carcinogenesis, carcinogenicity studies, epidemiological studies, evaluation of carcinogenicity of chemicals, carcinogenic substances and preparations.
  • 5. Reproduction and developmental toxicity, embryotoxicity, teratogenicity, testing of reproduction toxicity and teratogenicity, epidemiological studies, evaluation of reproduction toxicity, substances and preparations which are toxic for reproduction.
  • 6. Dangerous physical-chemical properties, testing and evaluation of dangerous physical-chemical properties of chemicals, explosive, oxidising, extremely flammable, highly flammable and flammable substances and preparations.
  • 7. Dangerous environmental properties of chemicals, ecotoxicology, testing and evaluation of ecotoxicity and other properties of chemicals, substances and preparations which are dangerous for environment.
  • 8. Factors determining the adverse effects of chemicals on organism - substance, organism, dose, the others.
  • 9. Interaction of substances with organism. Routes of exposure, absorption, distribution, biotransformation (reactions of the first and second phase of metabolic conversion) and elimination of xenobiotics, molecular, cellular and organ levels of interaction.
  • Exposure tests, biological threshold limit values, genetic risk monitoring of workers exposed to genotoxic agents.
  • 10. European Union legislation on substances and preparations - Directive No 67/548/EEC, Regulation No 1907/2006 (REACH) and No 1272/2008 (CLP).
  • Czech Republic legislation on substances and preparations, public health protection, occupational safety and health protection, and prevention of major accidents.
  • Permissible exposure levels and short-term exposure limits of chemicals and dusts in the air in the working zone, permissible concentrations of chemicals and dusts in indoor environment, maximum permissible concentrations of substances in drinking water, maximum residue levels in food.
  • 11. Risk assessment and occupation health protection of workpeople, individual protective devices, first aid measures in cases of accidental chemical exposure.
  • 12. Special toxicology of inorganic substances.
  • 13. Special toxicology of important organic compounds (aliphatic and aromatic hydrocarbons, halogenated hydrocarbons, alcohols, phenols, ethers, aldehydes, ketones, carboxylic acids and their derivatives, inorganic acid esters, nitro compounds, amines, organometallic compounds).
  • 14 Information sources on dangerous properties of substances and preparations. Safety data sheets, toxicological literature, databases on CD-ROM and on-line, toxicological information centre.
