Course objectives
The laboratory course aims to provide student with practical skills of methods of recombinant DNA technologies, genetic manipulation with bacteria and yeasts, expression of recombinant proteins in E. coli and P. pastoris, bacterial genome sequencing and transcriptomic and metagenomics analysis.
Learning outcomes
Student will have following practical skills: 1. Design and implement techniques related to DNA manipulation (cloning, transformation) 2. Design and perform expression of recombinant proteins in the E. coli and P. pastoris yeast expression system 3. To perform and evaluate metagenomics analysis from the environmental sample. 4. To perform and do a basic evaluation of bacterial genome and transcriptome sequencing data. 5. To perform critical evaluation of acquired data within basic methods of current gene technologies
Syllabus
  • Exercise 1 - Expression of Aspartate-tRNA Ligase Protein in E. coli BL21 (DE3) and its purification by affinity chromatography (IMAC) Exercise 2 - Expression of protein elicitin in P. pastoris GS115 and its subsequent purification by ion-exchange chromatography Exercise 3 - Metagenomics analysis of the environmental sample on the MiniSeq NGS instrument Exercise 4 - Analysis of bacterial transcriptome changes on the MiniSeq NGS instrument Exercise 5 - Bacterial genome sequencing on the MiniSeq NGS instrument Exercise 6 - Evaluation of acquired sequencing data.

Practical course
Assessment methods
After completing all exercises, a test consisting of 20 questions covering individual problems is written. To successfully complete the course, student must pass all the exercises and earns at least 70% of the test points.
