The relationship between obesity, diabetes mellitus type 2 (T2DM) and chronic periodontitis (CP), characterized by low-grade inflammation, was described. Adiponectin (ADIPOQ) regulates inflammatory responses and has multiple favorable effects on glucose and lipid metabolism. The aim of present study was to analyze ADIPOQ gene variability in healthy controls and patients with T2DM and/or CP. Altogether, 685 unrelated subjects were included in this case-control study. Genomic DNA of 104 healthy/non-periodontitis controls, 202 patients with CP and 380 T2DM patients (including 88 patients with T2DM and CP) were genotyped using the real-time PCR TaqMan method for +276G/T (rs1501299) polymorphism in the ADIPOQ gene. Clinical (diabetic complications, periodontal status, body mass index=BMI), biochemical (lipid profile, blood glucose and glycated hemoglobin levels) and microbial parameters (7 selected periodontal pathogens) were recorded. No association between ADIPOQ gene polymorphism and T2DM and/or CP was found (P>0.05). Subjects with TT genotype had significantly higher BMI and decreased median of high density lipoprotein (HDL) levels than in carriers of GT/GG genotypes (P≤0.01). However, no significant differences were found for triglycerides, total cholesterol, low-DL levels and other monitored parameters. Although the ADIPOQ gene variability +276G/T (rs1501299) was not associated with susceptibility to and severity of T2DM and/or CP in the Czech population, TT carriers may have a higher risk of obesity and dysregulation of lipid metabolism.
