Course objectives
At the end of this course, students should be able to work aseptically, to inoculate and cultivate bacteria, to prepare stained and native microscopic slides, to determine number of bacteria in different samples, to isolate bacterial group from chosen samples, to work with yeasts, to use some basic identification methods.
Syllabus
  • 1. Introduction, safety in microbiological laboratory 2. Media for cultivation and their preparation 3. Isolation, inoculation and cultivation of microorganisms 4. Microscopic examination of bacteria (slides preparation, Gram - staining, microscopy and image acquisition) 5. Determination of number of viable bacteria by plate count 6. Determination of the concentration of antibiotics by microbiological methods. 7. Determination of susceptibility of bacteria to antibiotics 8. Identification of bacteria - the use of biochemical tests and standardized Microsystems 9. Yeasts, test of vitality 10. Microorganisms in soil. 11. Isolation and microscopic examination of bacteria from soil (cellulolytic bacteria, Azotobacter sp., Clostridium sp.), negative - staining, capsule - staining. Microscopic examination of endospores. 12. Reports, test and microscopic examination/determination of unknown bacterial strain.

Teaching methods
Topic of each practical is at first explained and demonstrated to students. Then students are solving the practical task by themselves. The outcome of each practical is written report.
