Course objectives
At the end of the practical course of general microbiology students should be able to perform basic methods used in microbiological laboratory (methods of sterile inoculation and cultivation, slides preparation and microscopic examination of microorganisms, determination of the number of bacteria in different samples).
Learning outcomes
Outcomes of this course are students' protocols, which contain a short theoretical introduction, detailed methodology and results of individual practical tasks.
Syllabus
  • 1. Introduction, safety in microbiological laboratory 2. Media for cultivation and their preparation 3. Isolation, inoculation and cultivation of microorganisms 4. Microscopic examination of bacteria (slides preparation, Gram - staining, microscopy and image acquisition) 5. Determination of number of bacteria by cultivation method 6. Bacteria in water - basic microbiological analysis of water 7. Determination of the concentration of antibiotics by microbiological methods. Determination of susceptibility of bacteria to antibiotics 8. Identification of bacteria - use of biochemical tests and standardized Microsystems 9. Yeasts, test of vitality 10. Microorganisms in soil. Inoculation of microscopic fungi. 11. Isolation and microscopic examination of bacteria from soil (cellulolytic bacteria, Azotobacter sp., Clostridium sp.), negative - staining, capsule - staining. Microscopic examination of endospores. 12. Protocols, written test and identification of unknown bacterial strain by Gram staining.

Teaching methods
Topic of each practical is at first explained and demonstrated to students. Then students are solving the practical task by themselves. The outcome of each practical is written report.
Assessment methods
Active participation on practicals, submission of reports, written test and identification of an unknown bacterial strain by Gram staining.
