Course objectives
The main goal of the course is to enable students to understand the importance of individual analytical and biochemical techniques and to interpret their own experimental data, based on the techniques taught in the course. They should be able to work with the information in the primary literature, in order to understand and implement the techniques described in primary scientific literature in the area of animal physiology and cell biology. Based on this, they will be able to present and explain the techniques used in their own work to expert audience.
Learning outcomes
At the end of the course, students will be able to: identify and describe basic methodology for determination of protein levels and localization, estimation of protein-protein interactions, describe basic principles of manipulation of gene/protein expression and function; describe principles of isolation of nucleic acids and their analyses; describe methodology for analyses of lipids, saccharides and small organic molecules based on chromatography and mass spectrometry; describe basic cell culture techniques; describe omics technologies and basic methods of analytical cytometry; describe possibilities for application of animal models in cell biology and physiology;
Syllabus
  • 1) Tissue and cell culture, cultivation and analysis of cells; 2) Fluorescence techniques – fluorescence microscopy, flow cytometry, antibodies and their application in cell biology; 3) DNA isolation, separation techniques, PCR techniques, plasmid cloning and isolation, transfection of eukaryotic cells, manipulation of gene expression; 4) RNA isolation, detection (qRT-PCR, RNA-seq), non-coding RNAs, si/shRNA applications; 5) Protein analyses - separation techniques, Western blotting, immunochemistry, protein-protein interactions (immunoprecipitation); 6) Analyses of protein modifications, localization, proteomics (mass spectrometry), manipulation of protein level/activity; 7) Lipids and polysaccharides - isolation, separation and identification; 8) Separation and analytical techniques – HPLC, GC, mass spectrometry; 9) Omics techniques – genomics, proteomics, metabolomics, databases and large data sets; 10) Application of in vivo mammalian models in physiology and cell biology;
